RNA–protein interactions play a role in various cellular processes, and methods to study these are expanding.RNA–protein complexes can be isolated, after which bound proteins identified by western blot or mass spectrometry.Despite the variety of existing methods, it remains challenging characterize global interactome specific RNA transcript their native context upon perturbations.Promising technologies use RNA-targeting systems coupled with proximity-dependent biotinylation identify vivo. an important numerous processes health disease. In recent years, RNA-bound proteome has been extensively studied, uncovering many previously unknown RNA-binding proteins. However, little is known about particular bind transcript. this review, we provide overview interactions, focus on strategies that insights into transcripts. RNA interactome capture in yeast
RNA-binding proteins (RBPs) are key players in post-transcriptional regulation of gene expression in eukaryotic cells. To be able to unbiasedly identify RBPs in Saccharomyces cerevisiae, we developed a yeast RNA interactome capture protocol which employs RNA labeling, covalent UV crosslinking of RNA and proteins at 365nm wavelength (photoactivatable-ribonucleoside-enhanced crosslinking, PAR-CL)...
متن کاملNoncoding RNA transcripts.
Recent analyses of the human genome and available data about the other higher eukaryotic genomes have revealed that, in contrast to Eubacteria and Archaea, only a small fraction of the genetic material (ca 1.5%) codes for proteins. Most of genomic DNA and its RNA transcripts are involved in regulation of gene expression, which can be exerted at either the transcriptional level, controlling whet...
متن کاملLaser-mediated, site-specific inactivation of RNA transcripts.
The biological function of specific gene products often is determined experimentally by blocking their expression in an organism and observing the resulting phenotype. Chromophore-assisted laser inactivation using malachite green (MG)-tagged antibodies makes it possible to inactivate target proteins in a highly restricted manner, probing their temporally and spatially resolved functions. In thi...
متن کاملEnhancement of RNA Interference Effect in P19 EC Cells by an RNA-dependent RNA Polymerase
Background: RNA interference (RNAi) is a phenomenon uses double-stranded RNA (dsRNA) to specifically inhibit gene expression. The non-specific silencing caused by interferon response to dsRNA in mammalian cells limits the potential of utilizing RNAi to study gene function. Duplexes of 21-nucleotide short interfering dsRNA (siRNA) inhibit gene expression by RNAi. In some organisms, siRNA can als...
